[Ab] Resumo:OBJECTIVES: To explore the forensic application value of MPure-12 automatic nucleic acid purification (MPure-12 Method) for DNA extraction by extracting and typing DNA from bloodstains and various kinds of biological samples with different DNA contents. METHODS: Nine types of biological samples, such as bloodstains, semen stains, and saliva were collected. DNA were extracted using MPure-12 method and Chelex-100 method, followed by PCR amplification and electrophoresis for obtaining STR-profiles. RESULTS: The samples such as hair root, chutty, butt, muscular tissue, saliva stain, bloodstain and semen stain were typed successfully by MPure-12 method. Partial alleles were lacked in the samples of saliva, and the genotyping of contact swabs was unsatisfactory. Additional, all of the bloodstains (20 µL, 15 µL, 10 µL, 5 µL, 1 µL) showed good typing results using Chelex-100 method. But the loss of alleles occurred in 1 µL blood volume by MPure-12 method. CONCLUSIONS: MPure-12 method is suitable for DNA extraction of a certain concentration blood samples.Chelex-100 method may be better for the extraction of trace blood samples.This instrument used in nucleic acid extraction has the advantages of simplicity of operator, rapidity, high extraction efficiency, high rate of reportable STR-profiles and lower man-made pollution.

[Ab] Resumo:OBJECTIVES: To establish a method for rapid identification of bloodstain age. METHODS: Under laboratory conditions (20 ℃, 25 ℃ and 30 ℃), an integrating sphere ISR-240A was used as a reflection accessory on an UV-2450 UV-vis spectrophotometer, and a standard white board of BaSO4 was used as reference, the reflection spectrums of bloodstain from human ears' venous blood were measured at regular intervals. The reflection radios 541 and 577 at a specific wavelength were collected and the value of 541/ 577 was calculated. The linear fitting and regression analysis were done by SPSS 17.0. RESULTS: The results of regression analysis showed that ² of the ratios of bloodstain age to UV visible reflectivity in specific wavelengths were larger than 0.8 within 8 hours and under certain circumstances. The regression equation was established. The bloodstain age had significant correlation with the value of 541/ 577. CONCLUSIONS: The method of inspection is simple, rapid and nondestructive with a good reliability, and can be used to identify the bloodstain age within 8 hours elapsed-time standards under laboratory conditions.

[Ab] Resumo:In cases of firearm related fatalities a systematic investigation at the scene of death is indispensable to differentiate between self-inflicted and homicidal gunshot injuries. A common method to preserve gunshot residues (GSR) is their collection using adhesive tapes. However, the biological material gathered at the same time by the tapes would be of special interest if backspatter, ejected from the entrance wound against the direction of fire, could be detected. In the present study we examined the success rate of co-analysis of RNA and DNA recovered from biological traces sampled with adhesive tapes. The material originated from eight cases of fatal gunshots, taken from the hands of suspects or victims, examined 5 to 19 years ago for GSR. For all types of adhesive tapes tested, quantity and quality of the co-extracted nucleic acids was insufficient for successful DNA profiling, but was sufficient for the detection of blood-specific micro RNA (miRNA). In summary, sampling trace evidence from the hands of persons involved in fatal gunshots with adhesive tapes has a long-term detrimental effect on biological traces.

[Ab] Resumo:The study of blood stains retained at the scene of the crime is of crucial importance for the preliminary inquiry. The present article is focused on the analysis of the possibilities and prospects for the use of photogrammetry (PM) as exemplified by the foreign expert practice of the blood stains examination at the site of the event. It is shown that the results of the application of digital photogrammetry in addition to the traditional methods of morphological investigations enables the forensic medical experts to reconstruct a number of unique features and circumstances that accompanied the commission of a crime at the site of the event. Such PM techniques supplemented by the ballistic analysis of the blood splatter and droplet trajectories provides additional evidence that allows the forensic medical experts to reconstruct the scene of the crime including the pose and position of the victim at the moment of causing injury. Moreover, these data make it possible to determine the maximum number and the sequence of injurious impacts (blows). The authors discuss the advantages and relative disadvantages of the application of the photogrammetric technique in the routine practical expert work. It is emphasized that the published decision making algorithms provide the specialists in various disciplines and professional experts with the ready-made technological tools for obtaining the additional criteria for the objective improvement of the quality of the studies they carry out and for the enhancement of the value of expert conclusions. It is concluded that the application of the modern photogrammetric technologies can be recommended for the solution of the applied forensic medical problems and conducting the relevant expert research.

[Ab] Resumo:Luminol has been used for a long time for detecting latent blood traces during police investigations because it is easy to use and does not pose any health risks, while providing trace evidence for DNA analysis. It is often the method of choice for examining clothing. Clothes worn during the offense are often destroyed or washed afterwards by the offenders. The purpose of this study is to show the possibilities of blood and DNA detection on washed clothes by documenting the macroscopic results and their chemiluminescence after washing. The tests comprised different fabrics and laundry detergents including different washing and drying methods. Chemiluminescence was detected on almost all blood-marked samples (95.9%), even after all traces visible to the naked eye have been removed by washing. Evidence of a complete DNA profile or individual alleles could be confirmed in almost all of the test cases (93.3%).

[Ab] Resumo:Identification of human semen is indispensable for the investigation of sexual assaults. Fluorescence staining methods using commercial kits, such as the series of SPERM HY-LITER™ kits, have been useful to detect human sperm via strong fluorescence. These kits have been examined from various forensic aspects. However, because of a lack of evaluation methods, these studies did not provide objective, or quantitative, descriptions of the results nor clear criteria for the decisions reached. In addition, the variety of validations was considerably limited. In this study, we conducted more advanced validations of SPERM HY-LITER™ Express using our established image analysis method. Use of this method enabled objective and specific identification of fluorescent sperm's spots and quantitative comparisons of the sperm detection performance under complex experimental conditions. For body fluid mixtures, we examined interference with the fluorescence staining from other body fluid components. Effects of sample decomposition were simulated in high humidity and high temperature conditions. Semen with quite low sperm concentrations, such as azoospermia and oligospermia samples, represented the most challenging cases in application of the kit. Finally, the tolerance of the kit against various acidic and basic environments was analyzed. The validations herein provide useful information for the practical applications of the SPERM HY-LITER™ Express kit, which were previously unobtainable. Moreover, the versatility of our image analysis method toward various complex cases was demonstrated.

[Ab] Resumo:The spreading dynamics of blood dripping onto hard surfaces is compared to two spreading models. Samples of human blood, porcine blood, and Millipore water were dripped onto cardboard, foamcore, and glass surfaces in low velocity passive drip simulations. Final stain diameter, the total number of spines and scallops, and angle of impact were measured and analyzed. Spreading is best predicted by applying the concept of effective viscosity to the Scheller and Bousfield (R  = 0.91) and Roisman (R  = 0.89) spreading models. In the tested conditions, blood spreads with Newtonian tendencies; however, has quantifiable differences in stain appearance to Newtonian fluids like water. This is encouraging for the development of water-based fluids as synthetic blood substitutes (SBSs). The work presents an assessment platform to quantify and score the performance of simple water-based fluids using final stain diameter (6 points) and number of spines and scallops (6 points) at six dripping heights between 20 and 120 cm. The angle of impact of a stain alone is not a sensitive measure of SBS performance, but stain formation scores the SBS's performance with another 1 point. Together these features generate a quantitative relative ranking system, of a maximum possible 13 points, that can be used to support the use of a particular fluid for the creation of a drip stain. The performance of twenty simple fluids in the simulated dripping assessment test is described.

[Ab] Resumo:Raman spectroscopy is commonly used in chemistry to identify molecular structure. This technique is a nondestructive analysis and needs no sample preparation. Recently, Raman spectroscopy has been shown to be effective as a multipurpose analytical method for forensic applications. In the present study, blood identification and discrimination between human and nonhuman blood were performed by a portable Raman spectrometer, which can be used at a crime scene. To identify the blood and to discriminate between human and nonhuman blood, Raman spectra of bloodstains from 11 species (human, rat, mouse, cow, horse, sheep, pig, rabbit, cat, dog, and chicken) were taken using a portable Raman spectrometer. Raman peaks for blood (742, 1001, 1123, 1247, 1341, 1368, 1446, 1576, and 1619 cm ) could be observed by the portable Raman spectrometer in all 11 species, and the human bloodstain could be distinguished from the nonhuman ones by using a principal component analysis. This analysis can be performed on a bloodstain sample of at least 3 months old. The portable Raman spectrometer can be used at a crime scene, and this analysis is useful for forensic examination.

[Ab] Resumo:: In recent years, RNA analysis has been increasingly used in clinical and forensic genetics. Nevertheless, a major limitation of RNA-based applications is very low RNA stability in biological material, due to the RNAse activity. This highlights the need for improving the methods of RNA collection and storage. Technological approaches such as FTA Classic Cards (Whatman) could provide a solution for the problem of RNA degradation. However, different methods of RNA isolation from FTA cards could have diverse effects on RNA quantity and quality. The purpose of this research was to analyze the utility of three different methods of RNA isolation from peripheral blood collected on FTA Classic Cards (Whatman). The study also aimed at assessing RNA stability in bloodstains deposited on FTA cards. : The study was performed on peripheral bloodstains collected from 59 individuals on FTA Classic Cards (Whatman). RNA was isolated with High Pure RNA Isolation Kit (Roche Diagnostics), Universal RNA/miRNA Purification (EURx) and TRIzol Reagent (Life Technologies). RNA was subjected to quantitative analysis followed by reverse transcription and Real - Time PCR reaction. : The study has shown that FTA Classic Cards (Whatman) are useful tools for storing bloodstains at room temperature for RNA analysis. Moreover, the method of RNA extraction employing TRIzol Reagent (Life Technologies) provides the highest efficiency and reproducibility for samples stored for no more than 2 years. : The FTA cards are suitable for collecting and storing bloodstains for RNA analysis in clinical and forensic genetics.
