[Ab] Resumo:A taxonomic study was carried out on strain LW15 , which was isolated from the external lesions of diseased farmed Murray cod (Maccullochella peelii peelii) from an intensive culture pond. Cells of strain LW15 were Gram-negative, facultative-anaerobic, non-motile, and both coccobacillus- and bacillus-shaped. Growth was observed at NaCl concentrations of 0-2 % (w/v) (optimum, 0 %), 4-32 °C (optimum, 25-28 °C) and pH 5.0-9.0 (optimum, 7.0). Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain LW15 was affiliated to the genus Acinetobacter, showing the highest similarity to Acinetobacter guillouiae CIP 63.46 (97.7 %) and other Acinetobacter species with validly published names (93.5-97.6 %). Whole-genome sequencing and phylogeny reconstruction based on a core set of 1061 Acinetobacter genes indicated that strain LW15 was most closely related to the clade formed by A. guillouiae CIP 63.46 and Acinetobacter bereziniae CIP 70.12 and distantly related to any of the described species of genus Acinetobacter. Furthermore, strain LW15 could be distinguished from all known Acinetobacter species by its ability to assimilate ß-alanine and l-arginine, but not d-glucose. The principal fatty acids were C18 : 1ω9c, C16 : 0 and C16 : 1ω7c/C16 : 1ω6c. The major respiratory quinone was Q-9. Polar lipids of strain LW15 comprised diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, phosphatidylcholine, four phospholipids, aminolipid and two unknown lipids. Based on its phenotypic and genotypic data, strain LW15 represents a novel species of the genus Acinetobacter, for which the name Acinetobacterpiscicola sp. nov. is proposed. The type strain is LW15 (=MCCC 1K03337 =CICC 24241 =KCTC 62134 =JCM 32101 ).

[Ab] Resumo:Kudoa inornata is a myxosporean parasite that develops in the somatic muscle of spotted seatrout Cynoscion nebulosus, an economically and ecologically important fish in estuaries and harbors in southeastern North America. In South Carolina (SC), USA, over 90% of wild adult spotted seatrout are infected. To inform potential mitigation strategies, we conducted 3 experiments using naïve sentinel seatrout and infectious stages of K. inornata naturally present in raw water from Charleston Harbor, SC, to determine (1) if K. inornata infection follows a seasonal pattern, and (2) how long it takes for myxospores to develop in fish muscle. Infection by K. inornata was determined by visual detection of myxospores in fish muscle squashes, and any visually negative samples were then assayed for K. inornata ribosomal DNA using novel parasite-specific PCR primers. We observed that K. inornata infection in seatrout followed a seasonal pattern, with high prevalence when water temperature was highest (27-31°C; July-September) and infections that were either covert (at ~13-15°C) or not detected (<13°C) at the lowest water temperatures in January-February. Myxospore development occurred within 476 degree-days, i.e. 2 wk in a typical SC summer. Infection was dependent on fish density, which limited presumptive actinospore dose. Our findings suggest that the life cycle of the parasite may be disrupted by preventing spore-rich seatrout carcasses (e.g. at angler cleaning stations) being thrown back into harbors and estuaries throughout the year.

[Ab] Resumo:The present study focus the variation of carbon concentrations within three trophic level i.e., primary producer (phytoplankton), primary consumers (zooplankton) and secondary consumers (fish) in three selected ponds at East Kolkata Wetland area. Depending on the amount and frequency of wastewater input, physico-chemical characteristics of pond, species richness, predator-prey interactions and pond wise different piscicultural practices, the amount of carbon sequestration varied spatially. Significant temporal variations were also observed in each trophic level of these three selected East Kolkata Wetland pond ecosystems. On average primary producers were sequestered 2038.6 ± 244.8mg C m-3 d-1 whereas 307 ± 19.3 mg C m-3 and 11531.4 ± 318.2mg C m-3 was sequestered by primary and secondary consumers, respectively. In Kolkata and its nearby districts over 90% of the production was marked from the East Kolkata Wetland area. Consequently, a significant amount of sequestered carbon was exported from the East Kolkata Wetland ecosystem in the form of fish and this continuous system might increase the carbon sequestration efficiency of the aquatic ecosystem.

[Ab] Resumo:Spawning performance of pinfish Lagodon rhomboides without use of hormonal aids was monitored over an extended season. Nearly three million eggs were obtained from 75 spawns collected over a 90-day consecutive period from a single population of four brood fish (1M:1F). A mean ± s.d. batch fecundity of 30·27 ± 22·64 eggs g female was estimated with 98·0 ± 0·06% of the batch composed of floating eggs which were 1·04 ± 0·04 mm in diameter and 85·71 ± 27·59% fertile. Floating eggs successfully hatched 54·65 ± 29·13% of the time which yielded larvae that were 2·59 ± 0·24 mm in length. Fatty acids within floating eggs were largely represented by polyunsaturated fatty acids (45·30 ± 2·14% of total fatty acids) of which linoleic acid [(c18:2n-6cis) 3·49 ± 1·69% trifluoroacetic acid (TFA)] and docosahexaenoic acid (DHA) [(c22:6n-3) 28·47 ± 1·48% TFA] represented the majority of fatty acids for n-6 and n-3 polyunsaturated fatty acids, respectively. The strongest correlations between fatty acids and hatching success and larval survival to first feeding were observed for the DHA:EPA (eicosapentaenoic acid; c20:5n-3) ratio and total n-6 polyunsaturated fatty-acids levels, respectively. These data demonstrate potential for producers to rely on natural spawns for extensive egg production and provide a baseline for future development of natural spawning protocols of captive L. rhomboides.

[Ab] Resumo:The objective of this study was to investigate the optimal developmental time to perform sex reversal in Ussuri catfish Tachysurus ussuriensis, to develop monosex breeding in aquaculture. Systematic observations of gonadal sex differentiation of P. ussiriensis were conducted. The genital ridge formed at 9 days post fertilization (dpf) and germ cells begin to proliferate at 17 dpf. The ovarian cavity began forming on 21 dpf and completed by 25 dpf while presumptive testis remained quiescent. The primary oocytes were at the chromatin nucleolus stage by 30 dpf, the peri-nucleolus stage by 44 dpf and the cortical alveoli stage by 64 dpf. The germinal vesicle migrated towards the animal pole (polarization) at 120 dpf. In presumptive testis, germ cells entered into mitosis and blood vessels appeared in the proximal gonad on 30 dpf. The efferent duct anlage appeared on 36 dpf and formation of seminal lobules with spermatogonia and lobules interstitium occurred at 120 dpf. Therefore, gonadal sex differentiation occurred earlier in females than in males, with the histological differentiation preceding cytologic differentiation in T. ussuriensis. This indicates that undifferentiated gonads directly differentiate into ovary or testis between 17 and 21 dpf and artificial induction of sexual reversal by oral steroid administration must be conducted before 17 dpf.

[Ab] Resumo:Aquaculture has grown rapidly over the last three decades expanding at an average annual growth rate of 5.8% (2005-2014), down from 8.8% achieved between 1980 and 2010. The sector now produces 44% of total food fish production. Increasing demand and consumption from a growing global population are driving further expansion of both inland and marine aquaculture (i.e., mariculture, including marine species farmed on land). However, the growth of mariculture is dependent on the availability of suitable farming areas for new facilities, particularly for open farming practices that rely on the natural oceanic environmental parameters such as temperature, oxygen, chlorophyll etc. In this study, we estimated the marine areas within the exclusive economic zones of all countries that were suitable for potential open ocean mariculture activities. To this end, we quantify the environmental niche and inferred the global habitat suitability index (HSI) of the 102 most farmed marine species using four species distribution models. The average weighted HSI across the four models suggests that 72,000,000 km2 of ocean are to be environmentally suitable to farm one or more species. About 92% of the predicted area (66,000,000 km2) is environmentally suitable for farming finfish, 43% (31,000,000 km2) for molluscs and 54% (39,000,000 km2) for crustaceans. These predictions do not consider technological feasibility that can limit crustaceans farming in open waters. Suitable mariculture areas along the Atlantic coast of South America and West Africa appear to be most under-utilized for farming. Our results suggest that factors other than environmental considerations such as the lack of socio-economic and technological capacity, as well as aqua feed supply are currently limiting the potential for mariculture expansion in many areas.

[Ab] Resumo:The shortnose sturgeon (Acipenser brevirostrum LeSueur, 1818) is a vulnerable species that is found along the eastern coast of North America. Little is known about temperature tolerance in this species and with a rapidly changing global climate, it becomes increasingly important to define the thermal tolerance of this species to better predict population distribution. Using a modified critical thermal maximum test (CTMax), the objectives of this study were to determine the impact of heating rate (0.1, 0.2 and 0.25°Cmin ) on the thermal tolerance, associated hematological responses, and oxygen consumption in juvenile sturgeon. In addition, transcripts associated with physiological stress and heat shock (i.e., heat shock proteins) were also measured. Heating rate did not alter the CTMax values of shortnose sturgeon. Neither heating rate nor thermal stress affected plasma sodium and chloride levels, nor the expression of transcripts that included catalase, glucocorticoid receptor, heat shock protein70 (hsp70), heat shock protein 90α (hsp90α) and cytochrome P450 1a (cyp1a). However, regardless of heating rate, thermal stress increased both plasma potassium and lactate concentrations. Glucose levels were increased at heating rates of 0.2 and 0.25°Cmin , but not at 0.1°Cmin . Overall, oxygen consumption rates increased with thermal stress, but the response patterns were not affected by heating rate. These data support the hypothesis that shortnose sturgeon can tolerate acute heat stress, as many physiological and molecular parameters measured here were non-responsive to the thermal stress.

[Ab] Resumo:A novel virulent bacteriophage, vB_VspP_pVa5, infecting a strain of Vibrio splendidus was isolated from a sea-cage aquaculture farm in Greece, and characterized using microbiological methods and genomic analysis. Bacteriophage vB_VspP_pVa5 is a N4-like podovirus with an icosahedral head measuring 85 nm in length and a short non-contractile tail. The phage had a narrow host range infecting only the bacterial host, a latent period of 30 min and a burst size of 24 virions per infected bacterium. Its genome size was 78,145 bp and genomic analysis identified 107 densely-packed genes, 40 of which could be annotated. In addition to the very large virion encapsulated DNA-dependent RNA polymerase which is the signature of the N4-like genus, an interesting feature of the novel phage is the presence of a self-splicing group I intron in the thymidylate synthase gene. A tRNAStop interrupted by a ~2.5kb open reading frame-containing area was also identified. The absence of genes related to lysogeny along with the high efficacy observed during in vitro cell lysis trials, indicate that the vB_VspP_pVa5 is a potential candidate component in a bacteriophage cocktail suitable for the biological control of V. splendidus in aquaculture.

[Ab] Resumo:Abalone herpes-like virus (AbHV) and Red-spotted grouper nervous necrosis virus (RGNNV) are two serious viruses that infect animal populations in aquaculture. Both viruses cause diseases associated with high mortality rates, resulting in dramatic economic losses in the aquaculture industry. There are currently no effective treatments for either of these two viral diseases. Thus, early, rapid, and accurate diagnosis plays a fundamental role in disease prevention and control in aquaculture. Traditional methods of diagnosis, such as virus culture, enzyme-linked immunoassay, and polymerase chain reaction (PCR), are either time consuming or require sophisticated temperature control devices. In this study, one sets of specific primers and probes were designed for the real-time quantitative recombinase polymerase amplification (qRPA) detection of AbHV and RGNNV separately. The sensitivity and specificity of detection were evaluated by comparison with detection by conventional PCR and quantitative PCR. The optimal reaction temperature and time for virus detection is 37°C for 20min. The detection limit is 100 copies per reaction, making this approach faster and more sensitive than qPCR in this study. In a field application, the detection percentage of qRPA was higher than that of qPCR for both AbHV and NNV. Additionally, good correlation was found between qRPA and qPCR detection (R >0.8). The methods presented here can be used as alternatives to qPCR for quick and quantitative detection of pathogens infecting aquaculture species.
