[Ab] Resumo:In the work there are presented results of studies on the validation of the gas chromatographic (GC) methodfor the determination of chlorothalonil residue amounts in peaches with the use of electron capture detector (ECD). For the analytical control there was selected such stone fruit crop as the peach, referring to the crops, the most contaminated with residue amounts ofpesticides. There was justified the necessity of the inclusion in the procedure of the detection of the method of confirmation, based on mass spectrometry detection (MSD) (the type of ionization - electron impact). The significant source of the obtaining of incorrect data in the identification with the use of ECD of ions are shown to be phthalates, visualized in the chromatogram as intense and/or broad peaks. Mass spectra of compounds of the class ofphthalates are characterized by the dominant peak of the ion with the value of (mass/ charge) 149, just on this peak the detection of low molecular weight phthalates occurs in various matrices, on the spectrum there are also recorded typical ions corresponding to fragments of radical residues. The combination of the use of various types of the detection allows to prove that the revealed response (detector signal) is caused just by the analyte, but not the impurities, and optionally to optimize chromatographic conditions towards to the obtaining reliable results. The lower limit of the quantitation of chlorothalonil in peach fruits accounts for 0.01 mg/kg, determined with a signal/noise ratio of 10. The range of measured concentrations is volatile between 0.01-0.1 mg/kg, recovery rate of chlorothalonil from samples of peaches, established according to results of the analysis of model samples with the introduction of the substance in four points along the detection range, was 84-102%, the SD value of the repeatability varies in the range of 2.0-5.8%.

[Ab] Resumo:In the article there are presented results of a study of the balance of food consumption by the population of the Voronezh region, the assessment of the level of contamination of food products and both carcinogenic and non-carcinogenic and public health risk caused by this contamination. There was evaluated the level of nutrition-dependent diseases. A special feature of this study is a comparative analysis of data obtained during the two five-year periods of 1995-1999 and 2010-2014, that permitted to reveal changes in food consumption, the dynamics of morbidity rate associated with the nutritional factor.

[Ab] Resumo:In the article there is considered the quality control and safety system implemented in the one of the largest flight catering food production plant for airline passengers and flying squad. The system for the control was based on the Hazard Analysis And Critical Control Points (HACCP) principles and developed hygienic and antiepidemic measures. There is considered the identification of hazard factors at stages of the technical process. There are presented results of the analysis data of monitoring for 6 critical control points over the five-year period. The quality control and safety system permit to decline food contamination risk during acceptance, preparation and supplying of in-flight meal. There was proved the efficiency of the implemented system. There are determined further ways of harmonization and implementation for HACCP principles in the plant.

[Ab] Resumo:Food-borne infectious diseases remain an actual problem of health care. In the Republic of Belarus the level of food-borne diseases persists to be stable, at the same time these diseases remain to be significant. Despite the insignificant number of nonconforming food samples the prevalence rates of food-borne acute infectious diseases of various etiology with dominantly food transmission pathway remain to be rather high. The mentioned fact determines the need for the development of approaches on the security of microbiological food safety based on the risk analysis. There are determined criteria for a hygienic assessment of the microbiological hazards (associated with the pathogen, food products and a consumer health state), there is presented the classification offood products in dependence on the grade of microbiological risk. There was performed the comparative analysis of requirements for the microbiological safety of food accepted in the Republic of Belarus and European Economic Union, in the European Union and at the international level, which testify to the presence of separate differences. Based on the international approaches there was substantiated the multilevel control system of management of microbiological risks, there is reported the characteristic of separate criteria. There are determined the directions of the improvement of the security of microbiological safety of food.

[Ab] Resumo:A rapid, simple, and sensitive fluorescent immunochromatographic strip test (ICST) based on quantum dots (QDs) has been developed to detect 1-aminohydantoin (AHD), a major metabolite of nitrofurantoin in animal tissues. To achieve this, QD-labeled antibody conjugates, which consist of CdSe/ZnS QDs and monoclonal antibodies, were prepared by an activated ester method. Under optimal conditions, with the nitrophenyl derivative of AHD as the target, the ICST had a linear range from 0.1 to 100 ng/mL, with a correlation coefficient of 0.9656 and a 50% inhibitory concentration of 4.51 ng/mL. The limit of detection was 0.14 ng/g, which was below the minimum required performance limit of 1 µg/kg for AHD established by the European Commission. The recoveries for AHD ranged from 81.5% to 108.2%, with coefficients of variation below 13%, based on intraday and interday analysis. Furthermore, for AHD in real samples, the ICST showed high reliability and high correlation with liquid chromatography-tandem mass spectrometry (correlation coefficient of 0.9916). To the best of our knowledge, this is the first report of a novel and sensitive method based on a fluorescent ICST to detect AHD below the minimum required performance limit. The ICST demonstrated high reliability, and could be ideally suited for rapid, simple, and on-site screening of AHD contamination in animal tissues. [Ab] Resumo:Background: Rice is a staple food for many people in the world and an important ingredient for production of food for infants and young children. According to European Food Safety Authority (EFSA), cereals, primarily rice and rice products, are an important source of human exposure to inorganic arsenic, which has been classified by the International Agency for Research on Cancer (IARC) as group I carcinogen. Arsenic is present in rice and rice products mainly as an inorganic form being more toxic than organic compounds Objectives: The aim of the study was to determine the total and inorganic arsenic content in rice, rice-based products including food for infants and young children available on the market in Poland and thus to estimate consumer exposure to inorganic arsenic from these groups of foodstuffs Materials and Methods: A total of 62 samples of rice and rice products from trade, including a group of rice products for infants and young children, were tested. Contents of total and inorganic arsenic were determined by using hydride generation atomic absorption spectrometry (HGAAS), after dry mineralization of samples and reduction of arsenic to arsenic hydride with sodium borohydride. To extract the inorganic arsenic forms, the samples were subjected to hydrolysis in concentrated HCl and then reduced in the presence of hydrobromic acid and hydrazine sulphate after which triple chloroform extractions and triple 1M HCl re-extractions were performed. Exposure of different groups of populations (adults and children), was estimated in relation to the Benchmark Dose Lower Confidence Limit (BMDL05) as set by the Joint FAO/WHO Expert Committee on Food Additives (JECFA) that resulted in a 0.5% increase in lung cancer (3.0 µg/kg body weight (b.w.) per day) Results: Mean content of total and inorganic arsenic in investigated rice samples was 0.12 mg/kg (median: 0.09 mg/kg; 90th percentile 0.22 mg/kg) and 0.04 mg/kg (median: 0.03 mg/kg, 90th percentile 0.07 mg/kg). Brown rice was found to be more highly contaminated with both total and inorganic arsenic than white rice. Mean contamination of brown rice with total arsenic and inorganic arsenic was: 0.18 mg/kg (median: 0.12 mg/kg, 90th percentile: 0.32 mg/kg) and 0.05 mg/kg (median: 0.05 mg/kg, 90th percentile: 0.07 mg/kg). In turn for the white rice contamination was lower, mean total arsenic content: 0.10 mg/kg (median: 0.08 mg/kg, 90th percentile: 0.19 mg/kg) and mean inorganic arsenic: 0.03 mg/kg (median: 0.03 mg/kg, 90th percentile: 0.06 mg/kg). Contamination of rice-based products both total and inorganic arsenic was similar to those reported for rice, except rice wafers (mean: 0.24 mg/kg and 0.13 mg/kg). In the group of products for infants and young children obtained results were low ­ mean total arsenic content was 0.06 mg/kg and inorganic arsenic 0.02 mg/kg. The estimated average adult and children's exposure to inorganic arsenic with rice and rice products was less than 1% of the BMDL05. Intake of inorganic arsenic by 12-month-old infants with ricebased products intended for this group of population was at 6% BMDL05 Conclusions: Based on the obtained results, it was found that the content of total and inorganic arsenic in investigated samples of rice and rice products did not pose a health risk even though contamination levels in some individual samples were significant

[Ab] Resumo:Nanotechnology applications in the food industry, including food contact materials, offer many potential benefits for consumers and manufacturers alike. The article discusses the migration of nanoparticles from food contact materials and the possible health risks associated with in the context of insufficient knowledge of the potential exposure to nanomaterial. The importance of gaps in the general knowledge on the behaviour and biological interactions of nanomaterials in biological systems becomes crucial for risk assessment. The article also discussed numerous doubts concerning the measurements of biological reactions in animal tests and the need for new approaches in the interpretation of data from nanoparticles studies in vivo. The article underlines the need to develop predictive and validated toxicological tests that can be used to screen for potential hazards, and also to develop new methodology for measuring nanoparticles in biological matrices to assess human exposure. Further studies should focus on understanding the mechanisms of action. Nanoparticles exhibit chemical and physical properties that significantly differ from those substances at a large size. Different properties of nanoparticles may lead to different toxicological properties. From that reason nanoparticles, in each case, are individually assessed by the European Food Standard Agency (EFSA) in terms of health risk before the European Commission authorizes them to be used in food contact materials.

[Ab] Resumo:This study investigated the magnitude of heavy metal contamination and determined the carcinogenic as well as non-carcinogenic risks associated with selected food consumption in Bangladesh. Commonly consumed varieties of rice, vegetables, and fish samples were analyzed to measure the concentrations of heavy metals such as cadmium, chromium, lead, arsenic, manganese, nickel, and zinc. These staple food items showed the greatest probabilities of heavy metal contamination in different phases of their production and marketing. Wide variations of metal concentrations were observed. Specifically, estimated daily intakes of arsenic and cadmium exceeded allowable daily intakes in all three food items. Toxicity scores of the metals were evaluated, and a comprehensive risk assessment was conducted to quantify the risks associated with the daily food consumption. Except for cadmium and lead in vegetables, all the contaminants present in each food item posed significant levels of carcinogenic risks up to 2.99 × 10 compared to the EPA recommended carcinogenic risk level of 1.0 × 10 . Cadmium and arsenic intake due to rice consumption also posed unsafe levels of non-carcinogenic risks of 4.587 and 6.648, respectively, compared to the EPA recommended non-carcinogenic risk level of 1.0. Finally, a revised set of permissible limits was proposed for the heavy metals detected in the food items. Those permissible limits would ensure the risks associated with food consumption below the allowable carcinogenic and non-carcinogenic risk levels. Thus, this comprehensive approach would provide guidelines to formulate adequate control measures and regulatory limits of toxic metals in foods produced and marketed in Bangladesh.

[Ab] Resumo:Recently, magnetic solid-phase extraction has gained interest because it presents various operational advantages over classical solid-phase extraction. Furthermore, magnetic nanoparticles are easy to prepare, and various materials can be used in their synthesis. In the literature, there are only few studies on the determination of mycoestrogens in milk, although their carryover in milk has occurred. In this work, we wanted to develop the first (to the best of our knowledge) magnetic solid-phase extraction protocol for six mycoestrogens from milk, followed by liquid chromatography and tandem mass spectrometry analysis. Magnetic graphitized carbon black was chosen as the adsorbent, as this carbonaceous material, which is very different from the most diffuse graphene and carbon nanotubes, had already shown selectivity towards estrogenic compounds in milk. The graphitized carbon black was decorated with Fe O , which was confirmed by the characterization analyses. A milk deproteinization step was avoided, using only a suitable dilution in phosphate buffer as sample pretreatment. The overall process efficiency ranged between 52 and 102%, whereas the matrix effect considered as signal suppression was below 33% for all the analytes even at the lowest spiking level. The obtained method limits of quantification were below those of other published methods that employ classical solid-phase extraction protocols.

[Ab] Resumo:A simple, rapid UHPLC-MS/MS method has been developed and optimised for the quantitation of microcystins and nodularin in wide variety of sample matrices. Microcystin analogues targeted were MC-LR, MC-RR, MC-LA, MC-LY, MC-LF, LC-LW, MC-YR, MC-WR, [Asp3] MC-LR, [Dha7] MC-LR, MC-HilR and MC-HtyR. Optimisation studies were conducted to develop a simple, quick and efficient extraction protocol without the need for complex pre-analysis concentration procedures, together with a rapid sub 5min chromatographic separation of toxins in shellfish and algal supplement tablet powders, as well as water and cyanobacterial bloom samples. Validation studies were undertaken on each matrix-analyte combination to the full method performance characteristics following international guidelines. The method was found to be specific and linear over the full calibration range. Method sensitivity in terms of limits of detection, quantitation and reporting were found to be significantly improved in comparison to LC-UV methods and applicable to the analysis of each of the four matrices. Overall, acceptable recoveries were determined for each of the matrices studied, with associated precision and within-laboratory reproducibility well within expected guidance limits. Results from the formalised ruggedness analysis of all available cyanotoxins, showed that the method was robust for all parameters investigated. The results presented here show that the optimised LC-MS/MS method for cyanotoxins is fit for the purpose of detection and quantitation of a range of microcystins and nodularin in shellfish, algal supplement tablet powder, water and cyanobacteria. The method provides a valuable early warning tool for the rapid, routine extraction and analysis of natural waters, cyanobacterial blooms, algal powders, food supplements and shellfish tissues, enabling monitoring labs to supplement traditional microscopy techniques and report toxicity results within a short timeframe of sample receipt. The new method, now accredited to ISO17025 standard, is simple, quick, applicable to multiple matrices and is highly suitable for use as a routine, high-throughout, fast turnaround regulatory monitoring tool.
