Resumo:Two Gram-stain-positive, aerobic actinomycete strains, designated NEAU-PCY-3 and NEAU-PCY-4, were isolated from rhizosphere soil of Urtica urens L. collected from Anshan, Liaoning Province, northeast PR China. The 16S rRNA gene sequence analysis showed that the two strains exhibited 99.9 % 16S rRNA gene sequence similarity with each other and that they were most closely to Longispora fulva DSM 45356 (98.7, 98.9 %) and Longispora albida JCM 11711 (97.1, 97.2 %). Phylogenetic analysis based on the 16S rRNA gene sequences indicated that the two strains were located in the same lineage and formed a cluster with the genus Longispora. Both strains were observed to contain MK-10(H4) and MK-10(H6) as the predominant menaquinones. The cell wall peptidoglycan was found to contain meso-diaminopimelic acid, d-glutamic acid, glycine and l-alanine. Whole-cell hydrolysates mainly contained galactose, ribose and xylose. The phospholipid profile contained diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylinositol, several glycolipids and several unknown lipids. The major cellular fatty acids for strain NEAU-PCY-3 were iso-C16 : 0, iso-C17 : 0, anteiso-C17 : 0 and C18 : 1ω5c. The DNA-DNA hybridization value between strains NEAU-PCY-3 and NEAU-PCY-4 was 83.6±0.4 %, and the values between the two strains and their closest phylogenetic relatives, belonging to the genus Longispora, were well below 70 %, supporting that they represented a distinct genomic species. An array of phenotypic characteristics also differentiated the strains from their closely related species, the only two validly published Longispora species. On the basis of the genetic, chemotaxonomic and phenotypic properties, strains NEAU-PCY-3 and NEAU-PCY-4 were classified as representatives of a novel species of the genus Longispora, for which the name Longispora urticae sp. nov. is proposed. The type strain is NEAU-PCY-3 (=DSM 105119 =CCTCC AA 2017017 ).

Resumo:Three new sesquiterpene glycosides (1-3), three new glycerol glycosides (4-6), two new alkaloids (7-8), together with seven known compounds (9-15) all of which were isolated from the genus Pilea for the first time, were isolated from the whole plants of Pilea cavaleriei Levl subsp. cavaleriei. Their structures were determined by extensive spectroscopic techniques and chemical methods. The cytotoxic activity of the isolated compounds was evaluated against four cancer cell lines, and none of the tested compounds caused a significant reduction of the cell number.

Título:Antioxidant and antimicrobial activities of ethanol and aqueous extracts from Urtica urens.

Resumo:CONTEXT: Urtica urens L. (Urticaceae) is an important and commonly used plant for its medicinal and pharmacological properties. OBJECTIVE: We analyzed the antioxidant and antimicrobial activities of the leaves of Urtica urens in ethanol (EtOH) and water (WA) solvents, employing standard analytical methods. MATERIALS AND METHODS: Polyphenol, flavonoid and tannin content of Urtica urens leaves were determined, after their extraction, using EtOH (70%) and WA extracts as well as the antioxidant (DPPH, ABTS, ß-carotene and FRAP) and the antibacterial (via the method of dilution tests) activities of EtOH and WA extracts. RESULTS: The 70% EtOH of Urtica urens showed the highest values of total phenolic (31.41 mg GAE/g DW), flavonoids (6.81 mg quercetin/g DW), tannin (8.29 mg GAE/g DW) and TEAC (560 mmol Trolox/g DW), compared to the WA. The results of DPPH for EtOH (95.56%) were higher than that of WA (64.56%) at a concentration of 40 mg/L. The extracts displayed a FRAP 106.23 for EtOH and 30.55 µmol Fe(II)/g DW for WA. The results clearly indicated that EtOH was the strongest radical scavenger (IC = 245.65 ± 10.2 µg/mL). Ethanol was the most effective with minimum inhibitory concentration (MIC) < 250 µg/mL. WA has no antibacterial activity. DISCUSSION AND CONCLUSION: The results indicate that leaves of Urtica urens could be used as natural antioxidant and antimicrobial agents.

Resumo:OBJECTIVE: The aim of this study was to investigate the chemical composition and antioxidant properties of Urtica urens L.(Uu) as well as its anti-inflammatory effect on carrageenan (CARR)-induced paw oedema in rats. METHODS: The leaves were extracted using ethanol (EtOH) and water. The extracts were analysed for proximate composition and antioxidant activity using standard chemical analysis methods. RESULTS: The proximate analysis showed that Uu leaves contained appreciable percentages of dry mass, ashes, carbohydrates, proteins, lipids, sugars, anthocyanin, carotenoid and fibre. Results showed that EtOH extract contained the highest amount of total phenolics, flavonoids, tannins, ortho-diphenols and flavonols. It decreased the paw oedema after CARR administration, and ameliorated the activities of catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GPx), and the malondialdehyde (MDA). CONCLUSIONS: Uu displayed a high potential as a natural source of minerals, phytochemicals and antioxidant properties. EtOH extract exhibited a significant inhibition against CARR-induced inflammation.

Resumo:There is a well known link between inflammation and cancer during initiation, propagation and metastasis. Urera aurantiaca (UA) Wedd. (Urticaceae) is a medicinal plant used in traditional medicine to treat inflammatory processes with proven in vivo antiinflammatory and antinociceptive effects. The effects of a methanolic extract (UA) and a purified fraction (PF) on the proliferation of normal and tumoral lymphocytes under the effect of prostaglandin E (PGE ) and on nitric oxide production by lipopolysaccharide-stimulated macrophages was evaluated. Both UA and PF stimulated normal lymphocytes but, in presence of PGE a modulatory effect was observed. The normal lymphocyte proliferation induced by PGE was driven by pathways involving both PKC and H O . In macrophages, UA and PF did not modify cell viability and abrogated the synthesis of nitric oxide induced by lipopolysaccharide. In tumoral lymphocytes, the UA exerted a biphasic effect: at low concentrations it increased cell proliferation, while at high concentrations, it displayed an antiproliferative effect. UA and PF were capable of reverting the proliferative action of PGE . The tumoral cell proliferation induced by PGE is related to PKC, ERK 1/2 and MAP Kinase P38 pathways. The observed effects could be attributed to polyphenols, flavonoids and tannins. This work demonstrates the modulatory effects of the UA on different cell types during inflammatory conditions, which reinforces its antiinflammatory action. Copyright © 2016 John Wiley & Sons, Ltd.

Resumo:ETHNOPHARMACOLOGICAL RELEVANCE: Myrianthus arboreus P. Beauv (Cecropiaceae) is a medicinal plant distributed in forests and damp places of tropical Africa. Its leaves are widely used as food and/or for the treatment of various ailments including dysmenorrhoea, female infertility, tumors and diarrhea. However, to the best of our knowledge, no safety assessment of this plant has been reported yet. AIM OF STUDY: The present study aimed at evaluating the safety of the aqueous extract of leaves of Myrianthus arboreus (MAA) in Wistar rats through an acute and sub-acute oral administration. MATERIAL AND METHODS: In acute oral toxicity, the test was performed according to the Organization for Economic Cooperation and Development (OECD) guidelines Nr. 423 (acute toxicity class method, ATC) with slight modifications. Female Wistar rats were orally treated with the aqueous extract of M. arboreus at the doses of 2000 and 5000mg/kg. In sub-acute toxicity study, using the OECD guidelines Nr. 407, the extract was administered by gavage at the doses of 20, 110 and 200mg/kg/day for 28 consecutive days. RESULTS: A single oral administration of 2000 or 5000mg/kg of the extract induced neither mortality nor exterior signs of toxicity indicating a LD >5000mg/kg. In sub-acute study, the extract decreased triglycerides, total cholesterol/high density lipoproteins ratio and atherogenic index of plasma in both sexes at all tested doses. Alanine transaminase decreased in both sexes at 200mg/kg and serum creatinine levels decreased at all tested doses in females. Moreover, significant increases in ovarian and uterine wet weights, red blood cell count, hematocrit, mean corpuscular hemoglobin and hemoglobin were observed at 200mg/kg in females. In males, this extract decreased white blood cell count, lymphocytes and relative weight of seminal vesicles and ventral prostate at 200mg/kg. CONCLUSION: The aqueous extract of Myrianthus arboreus leaves was non-toxic in acute administration and exhibited a relatively low toxicity potential on accessory sex organs in both sexes, and leukocytes in males following the repeated 28-days oral administration of the dose 200mg/kg.

Resumo:The genus Urtica L. is subcosmopolitan, found on all continents (except Antarctica) and most extratropical islands and ranges from Alaska to Patagonia, Spitzbergen to the Cape and Camtschatka to the subantarctic islands. However, throughout its geographical range morphologically nearly indistinguishable species are found alongside morphologically quite disparate species, with the overall diversity of morphological characters extremely limited. The systematics of Urtica have puzzled scientists for the past 200years and no single comprehensive attempt at understanding infrageneric relationships has been published in the past, nor are species delimitations unequivocally established. We here provide the first comprehensive phylogeny of the genus including 61 of the 63 species recognized, represented by 144 ingroup accessions and 14 outgroup taxa. The markers ITS1-5.8S-ITS2, psbA-trnH intergenic spacer, trnL-trnF and trnS-trnG are used. The phylogeny is well resolved. The eastern Asian Zhengyia shennongensis T. Deng, D.G. Zhang & H. Sun is retrieved as sister to Urtica. Within Urtica, a clade comprising the western Eurasian species U. pilulifera L. and U. neubaueri Chrtek is sister to all other species of the genus. The phylogenetic analyses retrieve numerous well-supported clades, suggesting previously unsuspected relationships and implying that classically used taxonomic characters such as leaf morphology and growth habit are highly homoplasious. Species delimitation is problematical, and several accessions assigned to Urtica dioica L. (as subspecies) are retrieved in widely different places in the phylogeny. The genus seems to have undergone numerous dispersal-establishment events both between continents and onto different islands. Three recent species radiations are inferred, one in America centered in the Andes, one in New Zealand, and one in northern Eurasia which includes Urtica dioica s.str. sensu Henning et al. (2014). The present study provides the basis of a critical re-examination of species limits and taxonomy, but also of the dispersal ecology of this widespread plant group and an in-depth study of the three clades with recent radiations.

Resumo:Small auxin-up RNA (SAUR) genes are important gene families in auxin signalling transduction and are commonly used as early auxin responsive markers. Till date, no SAUR gene is identified in Urticales plants despite of the published bioinformation of mulberry, hemp and ramie. In this study, we used Arabidopsis sequences as query to search against mulberry, hemp genomes and ramie transcriptome database. In total, we obtained 62, 56 and 71 SAUR genes in mulberry, hemp and ramie, respectively. Phylogenetic analysis revealed the Urticales specific expansion of SAUR genes. Expression analysis showed 15 randomly selected ramie SAUR genes that were diversely functioned in ramie tissues and revealed a series of IAA-responsive, drought-responsive and high temperature-responsive genes. Moreover, comparison of qRT-PCR data and previous RNA-Seq data suggested the reliability of our work. In this study, we first report the identification of SAUR genes in Urticales plants. These results will provide a foundation for their function validation in Urticales plant growth and development.

Resumo:Natural plants products are one of the famous and commonly utilized remedies used in fighting diseases. This study was conducted to evaluate antioxidant activity of plants commonly used in Palestine (Urtica urens, Rumex cyprius and Borago officinalis). Free radical scavenging activity method was evaluated by using 2,2-diphenyl-1-picryl-hydrazyl-hydrate. The result show that the overall antioxidant activity of Rumex cyprius was the highest among the plants, followed by Urtica urens, and Borago officinalis; respectively. The (IC(50)) values of the methanolic extracts were 29.70±0.60µg/ml, 5.07±0.49µg/ml, 39.92± 0.52 µg/ml for Urtica urens, Rumex cyprius and Borago officinalis respectively. The results of this study revealed that these edible plants have high antioxidant activity and therefore they can provide natural sources of antioxidants and can be useful in preventing various diseases including cancer. These exhibited properties propose that such plants extracts can possibly be used as natural preservatives in the food and pharmaceutical industries and further characterization of Rumex cyprius constituents is needed.

Resumo:The natural carbon isotope composition of individual amino acids from plant leaf proteins has been measured to establish potential sources of variability. The plant leaves studied, taken from a range of plant groups (forbs, trees, grasses, and freshwater aquatic plants), showed no significant influence of either season or environment (water and light availability) on their Δδ(13)C values. Plant groups did, however, differ in carbon isotope composition, although no consistent differences were identified at the species level. A discriminant analysis model was constructed which allowed leaves from (1) nettles, (2) Pooideae, (3) other Poales, (4) trees and (5) freshwater higher plants to be distinguished from each other on the basis of their natural abundance (13)C/(12)C ratios of individual amino acids. Differences in carbon isotope composition are known to be retained, to some extent, in the tissues of their consumers, and hence an understanding of compound-specific variation in (13)C/(12)C fractional abundance in plants has the potential to provide dietary insights of value in archaeological and ecological studies.
